Title: Investigation of the Usage of Protein Copolymers, Obtained from Mediterranean Mussel Bisus, in Repair of Connective Tissues (Ligament/Tendon) and Suture Applications, while Preserving Their Native Properties

Abstract: In summary, single and multi-ply suture threads will be produced for the first-time using copolymer protein structures obtained by two different extraction methods from different parts of the mussel byssus. In addition, tissue scaffolds for screw-shaped ACL repair will be produced using a bioprinter for the first time using these copolymer structures. Whether the protein copolymer structures obtained by extraction preserve their self-healing properties will be examined on a molecular basis using the optical characterization method under polarized light. Fiber formation will be followed in situ by backscattering and Mueller Polarimetry methods. In the final stage of the project, the cytotoxic effects and cell viability of ACL scaffolds constructed with copolymer and collagen obtained from mussel bisus on tendon cells will be examined. In addition, the biocompatibility analysis of the new tissue scaffolds, their angiogenic potential will be tested by the chorioallantoic membrane (CAM) method.
This new scaffold raw material, which is planned to be developed by imitating nature at every stage of production (biomimetic), is thought to bring a new perspective, both in the field of materials science and medicine, provided that the properties of the raw material can be preserved (superior mechanical, self-healing, and surface adhesion) and transferred to newly produced materials.
